Preferred Specimen
Sputum from a single, first-morning,“deep-cough” sputum specimen, and the patient should not have eaten prior to collection.

Instructions
- Aseptically collect 2.0 mL of sputum in a specimen cup. The specimen should be a single, first-morning, “deep-cough” sputum specimen, and the patient should not have eaten prior to collection. When antibiotics are to be started immediately, a first-morning specimen is still preferred.
- Instruct patient to brush his/her teeth and/or rinse mouth well with water.
- Have patient remove dentures.
- Instruct the patient to take a deep breath, hold it momentarily, and cough deeply and vigorously into the container. Close lid securely. Submit specimen with CytoLyte®.
- If the patient is unable to produce sputum, a respiratory technician can induce the specimen. Induction may be effected by postural drainage, saline nebulization, or chest percussion.
- Label container (not the lid) with two patient identifiers (full name, DOB or MR), test requested, specimen source, and date and time of collection.
- Maintain sterility and forward promptly.
- Specimen source and clinical History/Diagnosis is required on request form for processing.
- Individual sputum specimens collected on 3 consecutive days are recommended.
- If cultures are desired, also submit an unpreserved specimen in a screw-capped, sterile container.
- NOCO Staff - Please send Cytology Order Requisition to Summit Pathology with specimen.
