Clinical Significance
The T-cell leukemia/lymphoma protein 1 (TCL1) is an AKT kinase regulator that is expressed in B cells up to the germinal stage and in plasmacytoid dendritic cells. TCL1 is strongly expressed, due to the chromosomal rearrangements, t(14;14)(q11;q32) and inv(14)(q11;q32), in 70% of cases of T-cell prolymphocytic leukemia (T-PLL) but is negative in all other mature T-cell lymphomas and leukemias. TCL1 is also expressed in nearly all cases of blastic plasmacytoid dendritic cell neoplasms, and can be used to distinguish this tumor from monocytic neoplasms. High TCL1 levels correlate with the pre-germinal center genetic subtype and are a strong adverse prognostic factor in B-cell chronic lymphocytic leukemia.
