PLATELET FUNCTION ASSAY (PFA-100)

Test Code
LAB318

CPT Codes
85576 x2

Includes
Collagen/Epinephrine test andCollagen/ADP test to detect platelet dysfunction by intrinsic platelet defects or exposure to medications containing platelet inhibiting agents.

Preferred Specimen
One 3 mL blue top tube.
Pediatric tubes are NOT ACCEPTABLE because the quantity of blood is insufficient for testing.

Instructions
Whole blood sample, do not centrifuge. Sample must be received within 3 hours of collection.
Sample must be collected by venipuncture (no butterfly or syringe), using a 21G or larger needle. Sample collected by IV/Line is NOT acceptable. A separate blue top tube MUST be collected for this test (ie, the collected tube CANNOT be used for other
testing. Tube MUST be filled to the etched line.

Transport Temperature
Ambient

Specimen Stability
Stability: Test must be performed within 4 hours of collection.

Reject Criteria (Eg, hemolysis? Lipemia? Thaw/Other?)
Clotted specimen, underfilled tube (not to etched line), sample drawn using a butterfly needle or collected through an IV/line, clumped platelets.  If sample platelet count is <75,000/uL, testing will not be performed in accordance with manufacturers guidelines. Samples with a low platelet count or hematocrit may have falsely elevated results.

Methodology
PFA-100 System

Setup Schedule
Sunday-Saturday

Reference Range
Collagen/Epinephrine: 82-150 seconds (Closure Time)
Collagen/ADP: 62-115 seconds (Closure Time)

Clinical Significance
Prolongation of PFA closure times (CT) is seen in congenital and acquired platelet function defects (PFD) and VWD. Anemia (Hct<28%)
and thrombocytopenia (Plt<100K/uL) can elevate PFA times. Prolonged Col/Epi CT with normal Col/ADP CT suggests aspirin induced
PFD. Prolongation of both Col/Epi and Col/ADP CT may suggest a PFD not related to aspirin.
