This test has been cleared or approved by the U.S. Food and Drug Administration and is used per manufactureer's instructions. Performance characteristics were verified by WVUH in a manner consisten with CLIA requirements.

Clinical Significance
DOG-1 IHC staining is primarily used to help identify gastrointestinal stromal tumors (GIST).

DOG-1 (discovered on GIST-1) functions in the regulation of cellular survival & proliferation as well as GI smooth muscle cholinergic activity.

DOG-1 IHC shows similar sensitivity and specificity for GIST as CD117 IHC, including those harboring KIT mutations (80-90%) as well as the
“wild-type” variant which lack KIT and PDGFRA mutations and are SDH-deficient. This latter tumor subtype is most common in the pediatric population. GIST with PDGFRA mutations (~10%) are typically negative for CD117 but are positive for DOG-1 in the majority of cases. Like CD117, DOG-1 is negative in schwannoma, leiomyoma/leiomyosarcoma, desmoid, melanoma and solitary fibrous tumor.

DOG-1 IHC is a cytoplasmic and membranous stain.
