Preferred Specimen
Two 2.7 ml (2.7 ml minimum) sodium citrate (blue) whole blood. Refer to Hemostasis/Thrombosis Specimens section for specimen collection instructions.


Minimum Volume
4.0 ml (1.5 ml minimum) citrated platelet-poor plasma


Instructions
Discontinue coumadin therapy for 14 days, direct thrombin inhibitors, fondaparinux and heparin 2 days prior to collection


Specimen Stability
Unspun and uncapped: 4 hours room temp; Separated from cells: 4 hours room temp, 2 weeks frozen plasma

Clinical Significance
The Prothrombin Time (PT) and Activated Partial Thromboplastin Time (APTT) mixing study is used to investigate
the cause of a prolonged PT or APTT screening result that is not due to the presence of heparin or coumadin in the
specimen. Once it is clearly established that the PT and/or APTT is prolonged, the task is to determine whether the
prolongation is caused by factor deficiency or by the presence of an inhibitor
