Preferred Specimen
1 mL platelet-poor plasma collected in a 3.2% sodium citrate (light blue-top) tube
Minimum Volume
0.5 mL plasma • 4.1 mL whole blood • 2.4 mL pediatric whole blood
Other Acceptable Specimens
5 mL (full tube) whole blood collected in 3.2% sodium citrate (light blue-top) tube • Pediatric: 2.7 mL whole blood collected in 3.2% sodium citrate (light blue-top) tube
Instructions
One full, unopened 3.2% sodium citrate tube (5 mL) whole blood. A completely filled tube is necessary because the correct ratio of blood to citrate is critical (9:1). Mix gentle inversion 3-4 times. Do not uncap.
Platelet poor plasma: Centrifuge light blue-top tube 15 minutes at approx. 1500g within 60 minutes of collection. Using a plastic pipette, remove plasma, taking care to avoid the WBC/ platelet buffy layer and place into a plastic vial/ Centrifuge a second time and transfer platelet-poor plasma into a new plastic vial/ Plasma must be free of platelets (<10,000/mcL). Freeze immediately and ship on dry ice.
***For Whole Blood there are Pediatric volumes- preferred volume: 2.7 mL Minimum volume: 2.4 mL. Transport temperature and stabilities same as adult volumes.****Draw plain red-top tube and discard or use for other testing. Draw blood into light blue-top tube, filling tube to mark indicated on label of tube. Do not overfill tube.

Clinical Significance
Screening test for abnormalities of coagulation factors that are involved in the extrinsic pathway. Also used to monitor effects of Warfarin therapy and to study patients with hereditary and acquired clotting disorders.
