Performed in the Microbiology Laboratory xX5785
Place sample into cup and add several drops of sterile non-bacteriostatic saline to keep tissue moist.
A single adequate sample may be used for multiple Microbiology requests.
 
*If anerobic culture is required, please add to requisition. Culture tissue will not automatically reflex to anaerobic culture.
Transport to Microbiology as soon as possbible  to insure successful anaerobic culture. it is crucial to minimize the period from time of collection to time of processing

Preferred Specimen
Surgical tissue, bone, biopsy, and other tissue specimens from areas which are sterile under normal conditions. Bone marrow aspirates and fine needle aspirates.


Minimum Volume
3-4 mm tissue or bone fragments.  Fine needle aspirate in a syringe with a luer lock cap, (needle removed).  
Bone marrow in pediatric isolator tubes (if specific  bacterial request. most often tested for fungal or AFB culture ) 


Instructions
For tissue: Collect biopsy sample from area within and adjacent to the area of infection. Avoid the necrotic tissue at the center of the infected area. Send sufficiently large sample (3-4 mm) to perform all of the microbiology tests. For fine needle aspirate: Insert the needle into the tissue, using various directions to draw up specimen, if possible. Remove the needle and submit with Luer-Lok on the syringe.

Sterile, screw cap cup, with sterile saline.
(Anaerobic swabs may be used in the OR, for anaerobic requests)

Do not refrigerate if anaerobic culture requested.


Reject Criteria (Eg, hemolysis? Lipemia? Thaw/Other?)
Swabs of tissue sites are the least desirable specimens for useful microbiology results. Swabs of tissue will be ordered as deep wound culture (CXDWD)

Swabs will be reordered as culture, Deeep wound (CXSWD) or culture, wound (CXWND) depending on the site


Reference Range
No Growth.


Clinical Significance
Healthy tissue sites are normally sterile,  except those such as colon biopsy expected to have normal flora.
Tissue specimens from decubitus ulcers, gangreous or necrotic tissue such as amputations or other wound debridements are commonly polymicrobial and also contain normal cutaneous flora
