Specimen Stability
Store specimens at room temperature (18 – 25oC), and process within 24 hours of collection.  


Reject Criteria (Eg, hemolysis? Lipemia? Thaw/Other?)


Grossly hemolyzed, QNS, or clotted




Methodology
Flow cytometry Stem Kit using CD34 antibodies

Setup Schedule
M-F only 9AM- 6PM.  
After 5 pm and on weekends, by prior arrangement only.
Call 6-5479 Flow Cytometry Lab.


Limitations


The presence of nucleated red blood cells, abnormal protein concentration, or hemoglobinopathies may result in unlysed red blood cells being counted as leukocytes,  and may cause falsely decreased CD34% results. The CD34+absolute counts should not be affected.  




Reference Range


Reference range for normal whole blood:
CD34+ (cells/μL): 0.50 – 6.50
CD34+ (%): 0.015 – 0.600 
 




Clinical Significance
Quantitate HPC (Human Progenitor Cells) in peripheral blood to predict ideal harvest time.
To quantitate HPC collected in apheresis product after harvest, to determine if further collections are needed.
To quantitate HPC in products for infusion (transplantation)
