Quantitative indirect immunofluorescence on the Phadia 250 (ThermoFisher Corp.)
Setup Schedule
This assay is performed twice per week.
Reference Range
MPO: <3.5 units –Negative
3.5 – 5.0 units - Equivocal
>5.0 units -Moderate to strong positive
PR3: <2.0 units –Negative
2.0 – 3.0 units - Equivocal
>3.0 units -Moderate to strong positive
3.5 – 5.0 units - Equivocal
>5.0 units -Moderate to strong positive

Clinical Significance
This test is used in conjunction with indirect immunofluorescence procedures to substantiate equivocal or positive pANCA or cANCA. The perinuclear staining pattern on neutrophils is most commonly associated with antibodies to myeloperoxidase. The cytoplasmic staining pattern on neutrophils is most commonly associated with antibodies to proteinase 3.
