Instructions
Material for eye cultures can usually be obtained by retracting the lower lid and stroking the conjunctiva with a sterile swab moistened with sterile saline. It is recommended that swabs for culture be taken before topical agents are applied. Drying of swabs should be avoided at all costs and the specimen should be transported to the laboratory as soon as possible. In some instances, corneal scrapings may provide evidence as to the nature of the disease. Because of the limited amount of material available, it is recommended that the scrapings be spot inoculated directly onto each of the following media available from the Microbiology laboratory: Chocolate agar, Sheep Blood agar, Sabouraud Dextrose agar and thioglycollate broth. The plates and broth should be brought to the Microbiology laboratory as soon as possible after collection. If acid fast organisms and/or fungi are sought, this should be brought to the attention of the Microbiology laboratory. If direct inoculaton of agar plates does not occur, the specimens should be collected using a culture swab or an eSwab. A Gram stain is NOT included in this culture.
