CHROMOSOME, HIGH RES

Message
"Pertinent medical findings must accompany request for chromosome analysis.
"

Test Code
52027

Alias/See Also
"High Resolution G-Banding
Prophase Analysis
"

CPT Codes
88230; 88262; 88289; 88291

Includes
"20 prophase to prometaphase cells are studied following cell synchronization. Two cells are karyotyped with the highest resolution.
"

Preferred Specimen
"Whole blood
10 mL (adult), 2 mL (pediatric)
"

Minimum Volume
"1 mL
"

Instructions
"Using sterile technique, collect 10 mL of blood into a sodium heparin tube or syringe. Invert the tube several times to prevent coagulation. Specimens must be sent the same day as collected and arrive in the laboratory within 48 hours of collection.
"

Transport Container
"Green-top (heparin) tube
"

Transport Temperature
room temp

Specimen Stability
"Specimen should be delivered immediately. Maintain specimen at room temperature. Specimen may be refrigerated if there is a delay in shipment. Do not freeze.
"

Reject Criteria (Eg, hemolysis? Lipemia? Thaw/Other?)
"Hemolysis; clotted specimen; use of improper anticoagulant; frozen specimen
"

Methodology
"Culture of cells in special medium; colcemid arrest of cells in prometaphase stage of mitosis following cell synchronization techniques "

Clinical Significance
"Detect small chromosome abnormalities not detectable using routine methods; precise identification of abnormal chromosomes previously detected by routine methods
"
