K-ras Pyrosequencing

Test Code
480250

CPT Codes
81275; 81403

Preferred Specimen
"Formalin-fixed, paraffin-embedded tissue (FFPE).
"

Minimum Volume
"2 mm x 2 mm tumor area
"

Instructions
"Maintain blocks and slides at room temperature.
Please provide four unstained slides and one H&E stained slide at 10 µM or tissue block.
Please provide a copy of the pathology report. Please direct any questions regarding this test to customer service at 800-345-4363.
"

Transport Container
"Slides or blocks
"

Reject Criteria (Eg, hemolysis? Lipemia? Thaw/Other?)
"Tumor block containing insufficient tumor tissue or tumor fixed in a heavy metal fixative; broken or stained slides
"

Methodology
"Polymerase chain reaction (PCR) and pyrosequencing "

Limitations
"DNA is isolated from a formalin-fixed, paraffin-embedded (FFPE) specimen and analyzed for mutations in the K-ras gene using PCR and pyrosequencing technologies. The methods used in this assay are highly sensitive, allowing the detection of approximately 5% of mutant in a background of wild-type genomic DNA.
This procedure may be considered by Medicare and other carriers as investigational and, therefore, may not be payable as a covered benefit for patients
