CULTURE, ACID FAST BACILLI (AFB/MYCOBACTERIA)

Alert the laboratory if Mycobacterium haemophilum, Mycobacterium ulcerans, Mycobacterium marinum, or Mycobacterium leprae is suspected Respiratory/Sputum: Collect 5 to 10 mL of respiratory specimen. Three sputum samples should be collected for acid fast smear and culture. These 3 samples should be collected at 8-24 hour intervals and include at least one first morning specimen. Submit in a sterile sealed leak proof container . Sterile Body Fluids and CSF: Submit in a sterile sealed leak proof container . Optimal volume is >5ml Urine: Obtain the entire first morning, clean-catch specimen when possible. Tissue and Biopsy Specimens: Submit in a sterile sealed leakproof container . Small biopsy samples should be moistened with a small (<1ml) amount of sterile saline for transportation Swab Samples: Submission of swab samples is strongly discouraged due to decreased recovery of Acid-Fast bacteria. Swabs are acceptable only if the specimen cannot be obtained by any other means. Specimens that leak in transit are not acceptable and will be rejected Immediate Specimen Processing: Specimens should be transported as soon as possible and must be received within 24 hours of collection (Refrigerated unless otherwise specified). For tissue specimens, a small amount of saline can be added to keep the tissue moist if necessary.
