COLLECTION OF SPUTUM SPECIMEN: PATIENT INSTRUCTIONS
Follow this procedure if the physician has requested an outpatient specimen collection for AFB culture, respiratory culture, or other testing on sputum specimens:
Collect specimen when patient first gets up in the morning, before eating:
Follow this procedure if the physician has requested an outpatient specimen collection for AFB culture, respiratory culture, or other testing on sputum specimens:
Collect specimen when patient first gets up in the morning, before eating:
- Brush teeth with a wet toothbrush, -- without toothpaste.
- Rinse mouth and gargle with water.
- Cough deeply from the lower chest and collect 5-10 ml sputum into the sterile container provided.
- Testing requires sputum specimen from a deep cough. Do not collect saliva (spit) from the mouth or postnasal discharge..
- Screw the lid carefully onto the container, and place the container inside the “zip-lock-type” bag which the lab provided. The bag has the word “Biohazard” on the outside of the bag.
- Bring specimen to the lab within 1-2 hours. If there is a delay of more than 3 hours, refrigerate the specimen. Specimen must be delivered to the lab before 2:00 pm on the day it is collected.

Clinical Significance
For cystic fibrosis respiratory specimens, Isolate and identify ALL potential pathogens, including sensitivity (even if just small quantity of pathogen). Major Pathogens include: Staphylococcus aureus, Haemophilus influenzae, Ps. aeruginosa, and Burkholderia cepacia complex.
