FEMALE ENDOCERVICAL Swab Specimen collection instructions:
  • Remove excess mucus from the cervical os and surrounding mucosa using the larger swab (white shaft). Discard that swab. Do NOT place this into the transport media tube.
  • Insert the specimen collection swab (blue shaft) into the endocervical canal. Gently rotate the swab clockwise for 10-30 seconds.
  • Withdraw the swab carefully-avoid contact with the vaginal mucosa.
  • Remove the cap from the transport tube and immediately place the specimen collection swab into the transport media.
  • Carefully break the swab shaft against the side of the transport tube. Discard the top portion of the shaft. Use care to avoid splashing of contents.
  • Re-Cap the transport tube tightly.
  • Label with patient information.
  • Forward to laboratory for testing.
  • GenProbe Aptima Vaginal Swab Specimen Collection Kit:   After Collection Storage:  Room or refrigerator temperature (2-300C).  Test within 60 days of collection.
  • SWAB SPECIMENS:  After collection, transport and store the swab in the swab specimen transport tube at  2°C to 30°C until tested

The Xpert Swab Specimen Collection Kit is used to collect, preserve, and transport swab specimens from these sources for assays.   

   

Specimen Collection Procedure:  Patient-collected Vaginal Swab

  1. Discard the larger cleaning swab in the package.
  2. Open the package that contains the pink-capped Xpert Swab Transport Reagent tube and individually wrapped collection swab.Set the tube aside before beginning to collect sample.
  3. Hold the swab in your hand, placing your thumb and forefinger in the middle of the swab shaft across the scoreline.
  4. Carefully insert the swab into your vagina about 5 cm (2 inches) inside the opening of the vagina and gently rotate the swab for 10-30 seconds.Ensure the swab touches the walls of the vagina so that moisture is absorbed by the swab.
  5. Withdraw the swab carefully.
  6. While holding the swab in the same hand, unscrew the pink cap from the Xpert Swab Transport Reagent tube.
  7. Take care to not spill the contents of the tube.If the contents spill on your skin, wash the affected area with soap and water.
  8. Immediately place the specimen collection swab into the transport reagent tube.
  9. Carefully break the swab shaft against the side of the tube at the scoreline and discard the top portion of the swab shaft.If needed, gently rotate the swab shaft to complete the breakage.Use care to avoid splashing contents on the skin.
  10. Re-cap the swab transport reagent tube and tighten the cap securely.
  11. Return the tube as instructed by your doctor, nurse, or care-provider who will then complete the following steps.
Steps for Caregiver to Perform
  1. Invert or gently shake the tube 3-4 times to mix the material from the swab.Avoid foaming.
  2. Label the transport tube with appropriate patient identification label, including date and time of collection, as required.
  3. Specimen source must be correct for the computer order.
  4. Transport and/or store the swab samples in the Xpert Swab Transport Reagent tube to the laboratory.
 
Clinician collected Endocervical Swab:
  1. Open the Xpert Swab Specimen collection kit.
  2. Before collecting the endocervical specimen with the Xpert Swab Specimen Collection Kit, remove excess mucus from the cervical os and surrounding mucosa using the large individually wrapped cleaning swab.Partially peel open the cleaning swab wrapper and remove the swab.Clean the cervical os and surrounding mucosa and then discard the swab.
  3. Open the package that contains the pink-capped Xpert Swab Transport Reagent tube and the individually wrapped collection swab.Set the tube aside before proceeding.
  4. Open the collection swab wrapper.
  5. Insert the collection swab into the endocervical canal.
  6. Gently rotate the swab clockwise for 10-30 seconds in the endocervical canal to ensure adequate sampling.
  7. Withdraw the swab carefully.
  8. While holding the swab in the same hand, unscrew the cap from the Xpert Swab Transport Reagent tube.
  9. Do not spill the contents of the tube.If the contents of the tube are spilled, use a new collection kit.If the contents spill on your skin, wash the affected area with soap and water.
  10. Immediately place the specimen collection swab into the transport reagent tube.
  11. Identify the scoreline on the collection swab shaft.Carefully break the swab shaft against the side of the tube at the scoreline and discard the top portion of the swab shaft.If needed, gently rotate the swab shaft to complete the breakage.Use care to avoid splashing contents on the skin.
  12. Re-cap the swab transport reagent tube and tighten the cap securely.
  13. Invert or gently shake the tube 3-4 times to mix the material from the swab with the reagent.Avoid foaming.
  14. Label the transport tube with appropriate patient identification label, including date and time of collection, as required.
  15. Transport and store the swab samples in the Xpert Swab Transport Reagent tube and send to the laboratory 
 
URINE SPECIMENS:
First Catch Male of Female Urine
  1. The patient should not have urinated for at least 1 hour prior to specimen collection.
    NOTE:  Catheter urine specimens are not acceptable for this test.
  2. Direct patient to provide a first-catch urine (approximately 20-30 ml of the initial urine stream).
  3. Female patients should not cleanse the labial area prior to providing the specimen.Male patients should not cleanse the tip of the penis before collecting.

QUICK REFERENCE SUPPLY GUIDE
  1. This Xpert Urine Specimen Collection Kit can be used on male or female urine and for Chlamydia trachomatis, Neisseria gonorrhoeae, and/or Trichomonas vaginalis.One urine sample is adequate for all 3 assays if requested.
  2. An aliquot of the patient’s specimen is pipetted into the yellow-capped Xpert Urine Transport Reagent.The disposable transfer pipette (individually wrapped) is provided with the kit.
  3. Fill to the black dashed fill line indicated on the tube.
  4. Invert the transport tube 3-4 times to ensure specimen and reagent are well mixed.
  5. Label the transport tube with appropriate patient identification label, including date and time of collection, as required.Do not obscure the black dashed fill line on the Urine Transport Reagent Tube.

Clinical Significance
The APTIMA Trichomonas vaginalis Assay is an in vitro qualitative nucleic acid amplification test (NAAT) for the detection of ribosomal RNA rRNA) from Trichomonas vaginalis to aid in the diagnosis of trichomoniasis using the PANTHER System.
The assay may be used to test the following specimens from symptomatic or asymptomatic women:
  • clinician-collected endocervical swabs,
  • clinician-collected vaginal swabs, and
  • Trichomonas vaginalis (TV) is the most common curable sexually transmitted disease (STD) agent in the United States.
    Infections in women cause vaginitis, urethritis, and cervicitis. Discharge and small hemorrhagic lesions may be present in the genitourinary tract. Complications can include premature labor, low-birth-weight offspring, premature rupture of membranes, and post-abortion or post-hysterectomy infection. An association with pelvic inflammatory disease, tubal infertility, and cervical cancer with previous episodes of trichomoniasis has been reported. Symptomatic women with trichomoniasis usually complain of vaginal discharge, vulvovaginal soreness, and/or irritation. Dysuria is also common. However, it has been estimated that 10 to 50% of T. vaginalis infections in women are asymptomatic, and in men the proportion may even be higher.
