Source Detailed Collection Information
Sputum
 
Sputum should be the result of a deep cough (not saliva) the first thing in the morning if practical or should be induced by an aqueous aerosol. Have patient remove dentures and rinse mouth with water. Collect 5 to 10 ml directly into a sterile container. Deliver to the laboratory within 1 hour. Refrigerate if can not be delivered immediately.
 
Tissue, Biopsy, or Bone
 
Collect tissue aseptically from the center and edge of the lesion.  Collect bone aseptically. Place specimen between moist gauze squares, add a small amount of sterile water or sterile non-bacteriostatic 0.85% saline to keep tissue from drying out, and send immediately to the laboratory.  Do not refrigerate.
 
CSF
 
Collect as much spinal fluid as possible, and place it in a sterile leakproof container. At least 1 mL is needed for Gram stain and culture. AFB and fungus culture require an additional minimum of 3 mL each. Generally, the no. 3 tube is used.  Do not refrigerate. Deliver to the laboratory immediately.
 
Body fluids (pleural,
synovial, and peritoneal)
Collect specimens aseptically, and place them in sterile leakproof containers. Do not refrigerate. Transport to the laboratory immediately.
 
Hair
 
Use a forceps to remove at least 10 infected hairs. Place hairs in a dry, sterile tube or container.
Nail
 
Remove nail polish from the nail to be sampled. Clean nail with 70% alcohol.
For a specimen of the dorsal plate, scrape the outer surface of the nail and discard the scrapings.  Then scrape the deeper affected portion of the nail to obtain the specimen.
Remove a portion of debris from under the nail with a scalpel.
The whole nail or nail clippings may also be submitted.
Place all material in a dry, sterile tube or container.
 
Skin and interspaces
 
Clean the skin surface with 70% alcohol. Scrape the surface of the skin at the active margins of the lesion(s) with a sterile scalpel. Place scrapings in a dry, sterile tube or container.
