Preferred Specimen
Specimen is produced by masturbation.
- The entire ejaculate must be collected in a clean screw-cap container provided by the laboratory or physician’s office
Patient Preparation
Patient should be abstinent for 2-3 days prior to collection. Provide patient with copy of “Instructions for collection of semen for analysis”.
Minimum Volume
A minimum volume of 1mL of semen is required.
Specimen Stability
Storage Temperature: Ideally, the specimen should be kept warm and brought to the lab immediately. However, any semen specimen is acceptable. Ask the patient when the specimen was collected and document in the computer.

Limitations
Method Limitations • Sperm may be missed if liquification is not complete or sample is not well-mixed. • This procedure is limited to post-vasectomy only. If semen analysis for fertility workup is ordered (to include sperm count, motility, etc), refer to closest reference lab that performs full semen analysis.

Clinical Significance
Post vasectomy semen checks are performed monthly to determine when sterilization is complete. A patient is considered sterile when two sperm checks, performed on samples collected a month apart, are negative for the presence of sperm cells.
