This test may reflex to CD68 IHC (per pathologist review) at an additonal charge (CPT code(s): 88342). The CD68 IHC will increase specificity for evaluating tumor-associated immune cells.


Instructions
Formalin-fixed, paraffin-embedded tissues are suitable for use.


Transport Container
Submit FFPE Block or 5 Unstained Slides. Unstained slides should be submitted on FLEX IHC Microscope slides or Fisherbrand Superfrost Plus charged slides. Unstained slides submitted must be sequential sections.

Submit Pathology report which includes the paraffin block number, tissue site, and both macroscopic and microscopic evaluation and diagnosis.

Note: Alternative fixatives may give erroneous results. Inconsistent results may be due to variations in fixation and embedding methods, or to inherent irregularities within the tissue

If sending precut slides, do not oven bake.

Ambient (Protect from excessive heat in summer with cold packs.)

6 months after sectioning


Reject Criteria (Eg, hemolysis? Lipemia? Thaw/Other?)
Depleted specimens with no tumor tissue remaining. Previously stained slides, Baked or Melted slides and Cytospin slides.

Clinical Significance
Immunotherapy has emerged as a promising treatment modality in cancer therapy. Immunohistochemistry (IHC) tests of PD-L1 expression may serve to select patients who will derive the most benefit from PD1- or PD-L1-directed therapy.

Additional CD68 staining may be required in certain cases to increase specificity. The CD68 antibody labels macrophages and other members of the mononuclear phagocyte lineage. This antibody is used to evaluate tumor-associated immune cells such as infiltrating macrophages.
