Clinical Significance
Haptoglobin is a plasma glycoprotein synthesized predominately by the liver. It consists of at least two heavy chains and two light chains covalently attached via disulfide linkage. Heterogeneity in molecular mass and structure is due to genetic polymorphism of the light chain where three major haptoglobin phenotypes are observed. A major role of haptoglobin is binding of hemoglobin in the bloodstream. Haptoglobin-hemoglobin complexes are transported to the liver or catabolic processing and preventing iron loss. The ability and capacity of haptoglobin to bind hemoglobin allows its use as an indication of intravascular hemolysis where levels are reduced or depleted. Haptoglobin is also an acute-phase protein where levels can exceed three times that of normal during an inflammatory response.
